Abstract

Aspects of the present disclosure include a complete genomic analysis device and methods of use thereof for high throughput genomic analysis of biological samples.

Core Innovation

The invention provides a complete genomic analysis device that includes a thermal chip module comprising multiple self-contained composite liquid cell (CLC) locations. A CLC production station is configured to access each self-contained CLC location, and the thermal chip module is part of a device architecture positioned on a main deck. The device further includes a sample receiving location and a reagent receiving location for handling liquids used in genomic analysis.

The device employs a robotically controlled liquid handler configured to transfer liquid between the sample receiving location, the reagent receiving location, and the thermal chip module. An interrogation station is configured to interrogate each self-contained CLC location of the thermal chip module. The thermal chip module, the CLC production station, the sample receiving location, the reagent receiving location, and the interrogation station are positioned on the main deck of the device, enabling operation as an integrated system for producing and interrogating CLC genomic reaction samples.

An egg sampling unit is operatively connected to the sample receiving location and configured to obtain biological samples from multiple avian eggs. In described embodiments, the egg sampling unit includes an avian egg tray, multiple egg receivers spaced at an egg pitch, and an egg sampling head comprising needles spaced at an egg pitch. The system integrates egg sampling with the thermal chip module and provides configurations for matching and aligning sampling positions with the CLC locations for genomic analysis and interrogation.

Claims Coverage

The independent claim defines 3 inventive feature groups: a deck-based genomic analysis device for producing and interrogating self-contained CLC locations on a thermal chip module, robotic liquid handling between sample and reagent locations, and an egg sampling unit for multiple avian eggs. Dependent claims refine the device with optical interrogation, cluster arrangements, and specific egg-sampling and mechanical connectivity features.

Complete genomic analysis device with deck-based thermal chip module and CLC production/interrogation

A complete genomic analysis device comprising a thermal chip module with multiple self-contained composite liquid cell (CLC) locations; a CLC production station configured to access each self-contained CLC location; a sample receiving location; a reagent receiving location; a robotically controlled liquid handler configured to transfer liquid between the sample receiving location, the reagent receiving location, and the thermal chip module; and an interrogation station configured to interrogate each self-contained CLC location, with the thermal chip module, CLC production station, sample receiving location, reagent receiving location, and interrogation station positioned on a main deck.

Egg sampling unit connected to sample receiving location for multiple avian eggs

An egg sampling unit operatively connected to the sample receiving location and configured to obtain biological samples from multiple avian eggs.

Optical interrogation of each self-contained CLC location using excitation and emission light

The interrogation station is configured to transmit excitation light to and collect emission light from each self-contained CLC location of the thermal chip module.

Thermal chip module configuration with multiple clusters and quantified CLC locations per cluster

The thermal chip module includes multiple clusters of self-contained CLC locations, and each multiple cluster comprises from 2 to 40 self-contained CLC locations.

Thermal chip module configuration with 84 clusters each having 17 self-contained CLC locations

The thermal chip module comprises 84 clusters each comprising 17 self-contained CLC locations.

Egg sampling unit with avian egg tray, egg receivers, and needle-spaced egg sampling head

The egg sampling unit comprises an avian egg tray, a plurality of egg receivers spaced at an egg pitch, and an egg sampling head comprising needles spaced at an egg pitch.

Mechanical connection allowing movement of thermal chip module to and from egg sampling unit

The thermal chip module is operatively connected to the egg sampling unit via a rail that permits movement of the thermal chip module to and from the egg sampling unit.

The inventive coverage combines a complete genomic analysis device that produces and interrogates self-contained CLC locations on a thermal chip module using robotic liquid handling and a deck-based layout, together with an egg sampling unit for multiple avian eggs connected to the sample receiving location. The dependent features further specify optical excitation/emission interrogation, quantified cluster-to-CLC location arrangements, and specific egg-sampling and mechanical connectivity features.
