Abstract

An object of the present invention is to find a novel substance that is capable of efficiently promoting the proliferation of stem cells while maintaining the undifferentiated state thereof and to provide the substance as an agent for maintaining the undifferentiated state of stem cells or an agent for promoting the proliferation of stem cells. Another object of the present invention is to provide an agent for treating a wound that has a wound healing effect on the skin and is readily available in the field of skin regenerative medicine or cosmetic treatment, and safe and inexpensive. The present invention relates to an agent for maintaining the undifferentiated state or promoting the proliferation of stem cells or an agent for treating a wound, comprising, as an active ingredient, one or a mixture of two or more of fatty acid glycerides represented by the following general formula (I): wherein at least one of R1, R2, and R3 represents an acyl group derived from an unsaturated fatty acid having 16 to 22 carbon atoms and the others each represent an acyl group derived from a saturated fatty acid or a hydrogen atom.

Core Innovation

The invention relates to stem cells and to promoting proliferation of stem cells. The invention provides a method in which stem cells are cultured in a medium comprising fatty acid diglycerides comprising at least one of 1-Palmitin-3-linolein, 1,3-Dilinolein, 1-Olein-3-linolein, 1-Stearin-3-linolein, 1,3-Diolein, and 1-Palmitin-2-linolein.

The invention further provides a method for promoting proliferation of stem cells in a mammal. In this method, an effective amount of fatty acid diglycerides is administered to the mammal, and the administered fatty acid diglycerides comprise at least one of the same listed diglycerides.

The invention also describes that mixtures of two or more of the specified fatty acid diglycerides are used. The document further includes undifferentiation and skin wound healing contexts in which fatty acid glycerides are used, along with reported results involving Nanog maintenance, increased proliferation, and improved in vitro scratch-wound recovery.

Claims Coverage

The provided relevant claims include two independent claims. Across these claims, the inventive subject matter centers on culturing stem cells in a medium containing specified fatty acid diglycerides and administering an effective amount of the same specified fatty acid diglycerides to a mammal to promote stem cell proliferation.

Culturing stem cells with specified fatty acid diglycerides

Culturing stem cells in a medium comprising fatty acid diglycerides comprising at least one of 1-Palmitin-3-linolein, 1,3-Dilinolein, 1-Olein-3-linolein, 1-Stearin-3-linolein, 1,3-Diolein, and 1-Palmitin-2-linolein.

Administering effective amount of specified fatty acid diglycerides to a mammal

Administering to the mammal an effective amount of fatty acid diglycerides comprising at least one of 1-Palmitin-3-linolein, 1,3-Dilinolein, 1-Olein-3-linolein, 1-Stearin-3-linolein, 1,3-Diolein, and 1-Palmitin-2-linolein.

Both independent claims require the use of fatty acid diglycerides selected from the listed set, with one claim directed to culturing in a medium and the other directed to administering an effective amount to a mammal; dependent claims further specify inclusion of at least one particular diglyceride, inclusion of at least two specified diglycerides, and use of an effective amount with mixtures of two or more diglycerides.

Stated Advantages

Efficient maintenance of the undifferentiated state (reported via Nanog maintenance).

Enhanced proliferation of stem cells (reported increase in proliferation, including in human somatic stem cells).

Effective wound healing (reported improved in vitro scratch-wound recovery).

Safe and inexpensive.

Documented Applications

Skin regenerative medicine / regenerative cosmetic treatment / treatment related to skin wound healing.

Use in undifferentiation and proliferation contexts for stem cells, including murine embryonic stem cells and human somatic stem cells (as reported in the document).

Use with product forms including cosmetic products and pharmaceutical products (and quasi drug), and food or drink (as described in the document).
