Abstract

The invention provides a method of preventing or reducing contamination of an object such as food with enterohaemorrhagic E. coli (EHEC), comprising contacting said object with colicin M or a derivative thereof.

Core Innovation

The invention relates to preventing or reducing contamination of an object with enterohaemorrhagic E. coli (EHEC) by contacting the object with colicin M or a derivative thereof in combination with one or more colicins selected from colicin Ia and colicin Ib. The colicin M and its derivative are defined by a translocation domain, a receptor binding domain, and an activity domain from the N- to the C-terminus.

The invention addresses the need to control EHEC contamination by employing colicin M due to its broad specificity described in the document. It targets EHEC pathogens associated with hemorrhagic colitis and hemolytic uremic syndrome (HUS) and enables reduction of EHEC contamination on objects, including foods and food objects.

The document further describes defined structural and sequence constraints for colicin M derivatives, including translocation domain residue ranges and allowable substitutions, insertions, additions and/or deletions compared to SEQ ID NO: 1. It also defines constraints for the central receptor-binding domain using residue ranges and allowable sequence identity or similarity, and describes aqueous compositions and freeze-dried preparations for contacting the object with colicin M or derivatives.

Claims Coverage

The partial content provides one independent claim. It covers a method of preventing or reducing EHEC contamination by contacting an object with colicin M or a derivative in combination with specified colicins, with additional coverage driven by domain architecture and derivative sequence constraints.

Contacting an object with colicin M in combination with colicins Ia and Ib

A method of preventing or reducing contamination of an object with enterohaemorrhagic E. coli (EHEC) by contacting the object with colicin M or a derivative thereof in combination with one or more colicins selected from the group consisting of colicin Ia and colicin Ib, wherein said colicin M and said derivative have a translocation domain, a receptor binding domain, and an activity domain from the N- to the C-terminus.

Derivative translocation domain constrained relative to SEQ ID NO: 1

The derivative of colicin M has a translocation domain of amino acid residues 1 to 35 of SEQ ID NO: 1 or a translocation domain having from 1 to 8 amino acid substitutions, insertions, additions and/or deletions compared to residues 1 to 35 of SEQ ID NO: 1.

Derivative central receptor-binding domain constrained by residues and identity/similarity

The derivative of colicin M comprises a central receptor-binding domain having residues 36 to 140 of colicin M of SEQ ID NO: 1, or having a domain with from 1 to 10 amino acid substitutions, insertions, additions and/or deletions compared to residues 36 to 140 of SEQ ID NO: 1, or having amino acid sequence identity of at least 90% to the receptor-binding domain of SEQ ID NO: 1, or having amino acid sequence similarity of at least 95% to the receptor-binding domain SEQ ID NO: 1.

Coverage centers on contacting an object with colicin M or a derivative plus specified colicins Ia and/or Ib, while enforcing an N-to-C domain architecture and specific residue, identity, and similarity constraints for the derivative translocation and central receptor-binding domains.

Stated Advantages
