Abstract

Provided herein is a nanoparticle comprising a metal core and a polymer shell coating the metal core useful as a magnetic resonance contrast agent.

Core Innovation

A nanoparticle is provided that includes a metal core and a polymer shell coating the metal core. The polymer shell comprises an inner shell and an outer shell, where the inner shell comprises a first polymer and the outer shell comprises a second polymer. The first polymer comprises PEG-PLA or PEG:PLGA, and the second polymer is PVP, PVA, a polyamide, or a combination thereof.

The nanoparticle is defined by a metal core to first polymer to second polymer mass ratio of about 1:0.25:3 to about 1:4:4. The polymer shell architecture with PEG-PLA or PEG:PLGA in the inner shell and PVP, PVA, or polyamide in the outer shell is used to support magnetic-resonance imaging contrast agent performance.

The described nanoparticles are used for MRI diagnostic imaging, including binding to amyloid plaques for neurodegenerative diseases such as Alzheimer’s disease. The described approach further includes in vivo MRI labeling of amyloid-beta plaques and use as a pharmaceutical composition administered parenterally.

Claims Coverage

The independent claim defines a nanoparticle with a metal core and a two-layer polymer shell, with an inner PEG-PLA or PEG:PLGA shell and an outer PVP, PVA, polyamide, or combination thereof shell, and a specified metal-core/first-polymer/second-polymer mass ratio window of about 1:0.25:3 to about 1:4:4.

Two-layer polymer shell with PEG-PLA/PEG:PLGA inner shell and PVP/PVA/polyamide outer shell

A nanoparticle comprising a metal core and a polymer shell coating the metal core, where the polymer shell comprises an inner shell comprising a first polymer and an outer shell comprising a second polymer, wherein the first polymer comprises PEG-PLA or PEG:PLGA and the second polymer is PVP, PVA, a polyamide, or a combination thereof.

Metal core to first polymer to second polymer mass ratio window

The mass ratio of the metal core to the first polymer to the second polymer is about 1:0.25:3 to about 1:4:4.

Overall, the claim coverage centers on the specific two-layer polymer shell composition and the defined mass ratio range linking the metal core with both polymer layers.

Stated Advantages

Reduced agglomeration.

Improved long-term stability and circulation.

Improved blood-brain barrier penetration.

Documented Applications

MRI diagnostic imaging using the nanoparticle as an MRI contrast agent, including amyloid plaque binding for neurodegenerative diseases such as Alzheimer’s disease.

In vivo MRI labeling of amyloid-beta plaques in a mouse imaging context (5×FAD is explicitly referenced in the provided content).

Use as a pharmaceutical composition for MRI diagnostic imaging administered parenterally.
