Abstract

Disclosed are products and methods for treating soft tissue injuries. The provided methods include the production of processed or cryopreserved microvascular tissue. Also provided are products and methods of using processed or cryopreserved microvascular tissue for the treatment of soft tissue injuries.

Core Innovation

The invention relates to a processed microvascular tissue that comprises a dried microvascular tissue, wherein the microvascular tissue comprises dissociated and uncultured stem or progenitor cells having intact cell membranes. The viability of the uncultured stem or progenitor cells in the processed microvascular tissue is less than 30%, while the processed microvascular tissue has tissue healing activity. The processed microvascular tissue is presented as dried tissue produced by drying techniques including freeze-drying or spray-drying.

The invention describes cryopreserved microvascular tissue comprising a frozen microvascular tissue and an excipient. The cryopreserved microvascular tissue comprises dissociated and uncultured stem or progenitor cells having intact cell membranes, with a viability of the uncultured stem or progenitor cells in the cryopreserved microvascular tissue being less than 30%, and the cryopreserved microvascular tissue having tissue healing activity when thawed. The excipients and processing formats are described as enabling processed microvascular tissue to retain tissue healing activity despite low viability and/or non-viable cells.

The invention addresses soft-tissue repair by applying processed microvascular tissue to a tissue selected from the group consisting of tendon, ligament, and skin to provide improved repair or regeneration compared with a control tissue. The invention further discusses optional hard-tissue healing as being part of the described tissue healing activity, including improved healing for soft or hard tissue exposed to the composition compared with analogous tissue treated without exposure to the composition.

Claims Coverage

The provided independent claims are directed to two core compositions—(i) a dried processed microvascular tissue and (ii) a cryopreserved microvascular tissue—each defined by dissociated, uncultured stem/progenitor cells with intact membranes, low viability (<30%), and tissue healing activity, including tissue healing upon thawing for the cryopreserved form. Across the dependent claim refinements, the claims add quantitative viability thresholds, specify component sets (e.g., excipients and optional matrix/clot-related components), and define repair/regeneration uses for tendon, ligament, and skin with improved healing versus control.

Dried processed microvascular tissue with intact membranes and low viability

A processed microvascular tissue comprising a dried microvascular tissue, wherein the microvascular tissue comprises dissociated and uncultured stem or progenitor cells having intact cell membranes, wherein the viability of the uncultured stem or progenitor cells in the processed microvascular tissue is less than 30%, and wherein the processed microvascular tissue has tissue healing activity.

Cryopreserved microvascular tissue with excipient and thawed tissue healing activity

A cryopreserved microvascular tissue comprising a frozen microvascular tissue and an excipient, wherein the microvascular tissue comprises dissociated and uncultured stem or progenitor cells having intact cell membranes, wherein the viability of the uncultured stem or progenitor cells in the cryopreserved microvascular tissue is less than 30%, and wherein when thawed, the cryopreserved microvascular tissue has tissue healing activity.

Together, the independent claims cover dried or cryopreserved microvascular tissue compositions containing dissociated, uncultured stem or progenitor cells with intact cell membranes and low viability (<30%) that nonetheless exhibit tissue healing activity; the cryopreserved form specifies retention of tissue healing activity upon thawing. Dependent claim refinements further narrow viability thresholds and enumerate optional excipients and additional components, and define comparative repair/regeneration outcomes for tendon, ligament, and skin versus control.

Stated Advantages

Improved tissue healing using a processed microvascular tissue comprising low-viability (<30%) dissociated uncultured stem or progenitor cells with intact cell membranes.

Cryopreserved microvascular tissue retains tissue healing activity when thawed despite low viability (<30%).

Improved repair or regeneration compared with an untreated control tissue when applied to tendon, ligament, and/or skin.

Documented Applications

Repairs or regenerates a tissue selected from the group consisting of tendon, ligament, or skin by applying a composition containing the processed microvascular tissue.

Improved healing activity is described for soft or hard tissue exposed to the composition compared with analogous tissue similarly treated but without exposure to the composition.
