Abstract

The present invention relates to a pharmaceutical composition for preventing, treating or delaying an Alzheimer's disease (AD) or dementia including a G protein-coupled receptor19 (GPCR19) agonist or its pharmaceutically acceptable salt as an active ingredient and a food composition for preventing, treating or delaying an Alzheimer's disease or dementia. The GPCR19 agonist according to the present invention has an effect of improving cognitive and behavioral disorders without harming the health of objects when administrated to the objects and has an effect of preventing the Alzheimer's disease or dementia or delaying or treating the progression of the disease in the objects with the disease by suppressing apoptosis of the brain tissue, enhancing immunity, and reducing formation of an amyloid β (Aβ) plague.

Core Innovation

The disclosed subject matter provides pharmaceutical and food compositions for preventing, treating, or delaying Alzheimer’s disease or dementia by using a G protein-coupled receptor19 (GPCR19) agonist as an active ingredient. The GPCR19 agonist is sodium taurodeoxycholate, including derivatives and pharmaceutically acceptable salts, and the approach is directed to an individual having Alzheimer’s disease or dementia via administration of a pharmaceutically effective amount.

The described rationale links GPCR19 agonist treatment with suppression of brain apoptosis and changes associated with neuroinflammation and amyloid pathology. The disclosure reports enhanced immunity, suppression of apoptosis, reduced amyloid β plaque formation, and reductions in microglia and astrocyte inflammatory markers, including iNOS and ROS, together with a decrease in apoptotic indicators in the brain.

In reported results, treatment is associated with improved cognitive and behavioral outcomes in a 5xFAD mouse model, together with observed blood changes. The disclosure states that animals do not show harm, with no adverse weight change, and describes improved Morris water maze performance and probe measures, increased CCL3, and decreased glucose.

Claims Coverage

The independent claim covers treating Alzheimer’s disease or dementia by administering a pharmaceutical composition containing a pharmaceutically effective amount of a GPCR19 agonist, specifically sodium taurodeoxycholate, to an individual with Alzheimer’s disease or dementia. Dependent claims further refine the treatment by specifying permitted administration routes, daily dose range constraints, and dosing frequency.

GPCR19 agonist sodium taurodeoxycholate pharmaceutical treatment of Alzheimer’s disease or dementia

Administering to an individual having Alzheimer's disease or dementia a pharmaceutical composition containing a pharmaceutically effective amount of a G protein-coupled receptor19 (GPCR19) agonist, wherein the GPCR19 agonist is sodium taurodeoxycholate.

Permitted administration routes for the sodium taurodeoxycholate pharmaceutical composition

Administering the pharmaceutical composition formulated for administration by one or more of: oral, rectal, intravenous, intramuscular, intraperitoneal, subcutaneous, intranasal, or percutaneous.

Daily dose range for GPCR19 agonist sodium taurodeoxycholate

Administering a daily dose of the GPCR19 agonist ranging from 0.1 mg/kg/day to 1000 mg/kg/day.

Intermediate daily dose subrange for sodium taurodeoxycholate

Administering a daily dose of the GPCR19 agonist between 1 mg/kg/day and 500 mg/kg/day.

Weekly dosing frequency for GPCR19 agonist sodium taurodeoxycholate

Administering the GPCR19 agonist 1 to 3 times per week.

Overall claim coverage centers on administering sodium taurodeoxycholate, a GPCR19 agonist, in a pharmaceutically effective amount to treat Alzheimer’s disease or dementia, with dependent claim refinements regarding administration routes, daily dose range, and dosing frequency.
