Abstract

The present invention relates to iota- and/or kappa-carrageenan in combination with a neuraminidase inhibitor for use as a medicament in the prophylactic or therapeutic treatment of a symptom, condition or disease caused by or associated with an infection by an influenza virus.

Core Innovation

The invention provides a pharmaceutical composition for the treatment of a symptom, condition or disease caused by an infection with influenza virus or by a co-infection with influenza virus and at least one other respiratory virus. The condition or disease includes acute bronchitis, chronic bronchitis, rhinitis, sinusitis, croup, acute bronchiolitis, pharyngitis, tonsillitis, laryngitis, tracheitis, asthma and pneumonia, and the symptom includes fever, pain, dizziness, shivering, sweating and dehydration. The composition comprises an antiviral effective amount of at least one carrageenan selected from iota carrageenan and kappa carrageenan, and an antiviral effective amount of zanamivir as a neuraminidase inhibitor.

The approach addresses the background problem that antiviral treatment with a neuraminidase inhibitor has a limited treatment window, while carrageenan antiviral activity is associated with respiratory mucosa. The invention positions a synergistic combination to improve treatment of influenza virus infection and influenza co-infections. The disclosed rationale includes that carrageenan antiviral activity at respiratory mucosa can be combined with neuraminidase inhibitor activity.

The disclosed embodiments include iota and/or kappa carrageenan combined with a neuraminidase inhibitor, including zanamivir or oseltamivir. Partial content further discloses PCR evidence of influenza co-infections with other respiratory viruses including rhinovirus, coronavirus, and paramyxovirus, supporting treatment for infection with influenza virus and co-infection. Documented experimental disclosures in a mouse model report improved survival and symptom proxies such as weight loss for combination therapy versus monotherapies when treatment starts 24 h or 48 h post infection, including a triple combination of iota and kappa carrageenan with zanamivir.

Claims Coverage

The independent claim disclosed in the provided claims is directed to a pharmaceutical composition for treating influenza-related symptoms and conditions, including influenza co-infections with at least one other respiratory virus. The independent claim requires a dual antiviral combination of carrageenan, iota and/or kappa, with zanamivir as a neuraminidase inhibitor, with specified concentrations.

Influenza symptom and condition treatment with carrageenan plus zanamivir at specified concentrations

A pharmaceutical composition for treatment of a symptom, condition or disease caused by an infection with influenza virus or by a co-infection with influenza virus and at least one other respiratory virus, where the condition or disease is selected from acute bronchitis, chronic bronchitis, rhinitis, sinusitis, croup, acute bronchiolitis, pharyngitis, tonsillitis, laryngitis, tracheitis, asthma and pneumonia, and the symptom is selected from fever, pain, dizziness, shivering, sweating and dehydration, comprising an antiviral effective amount of at least one carrageenan selected from iota carrageenan and kappa carrageenan, and an antiviral effective amount of zanamivir as a neuraminidase inhibitor, wherein the concentration of carrageenan is 1.2 mg/ml and the concentration of zanamivir is 0.1 mg/ml in the composition.

The core claim coverage centers on treating influenza virus infection and influenza co-infection using an antiviral effective amount of iota and/or kappa carrageenan combined with zanamivir as a neuraminidase inhibitor, with the carrageenan concentration of 1.2 mg/ml and the zanamivir concentration of 0.1 mg/ml.

Stated Advantages

The combination is positioned to improve treatment of influenza virus infection and influenza co-infections.

Mouse model disclosures report improved survival and symptom proxies such as weight loss for combination therapy versus monotherapies.

The disclosed rationale includes that carrageenan antiviral activity at respiratory mucosa can be combined with neuraminidase inhibitor activity.

Documented Applications

Treatment of symptoms, conditions or disease caused by an infection with influenza virus or by a co-infection with influenza virus and at least one other respiratory virus, including acute bronchitis, chronic bronchitis, rhinitis, sinusitis, croup, acute bronchiolitis, pharyngitis, tonsillitis, laryngitis, tracheitis, asthma and pneumonia; and symptoms including fever, pain, dizziness, shivering, sweating and dehydration.

Treatment for influenza co-infection involving other respiratory viruses including rhinovirus, coronavirus and paramyxovirus.

Mouse model disclosures reporting improved survival and symptom proxies such as weight loss for combination therapy versus monotherapies when treatment starts 24 h or 48 h post infection.
